A transmission electron microscope (TEM) is a type of electron microscope that produces high-resolution images of thin specimens by transmitting a beam of electrons through them. The specimen is held on the end of a metal wire or rod, which is inserted into the TEM's vacuum chamber. A voltage difference between the specimen and the cathode (the negatively charged part of the electron gun) creates an electric field that accelerates the electrons passing through the specimen. These accelerated electrons interact with atoms in the sample, causing them to emit secondary electrons. A series of lenses inside the TEM collects these secondary electrons and focuses them onto an image detector, usually a phosphor screen or photographic film.
