ELISA is an abbreviation of Enzyme-linked immunosorbent assay. ELISA is a technique to measure the quantity of antigens or antibodies in a sample. The sandwich ELISA method consists of two antibody-coated surfaces, one surface coated with antigen and the other surface coated with antibody specific for the antigen type. The sandwich ELISA is also known as the "sandwich immunoassay" or "sandwich assay". The sandwich ELISA method was first described by Voller in 1975. The principle of this technique is to allow free antigen-antibody reaction between the microtiter plate wells coated with antibody, and then add a second antibody that has been linked to an enzyme. After an incubation period, a chromogenic substrate will be added to the wells where bound enzyme reacts with it. This causes a change in color that can be measured spectrophotometrically at 405 nm or 490 nm depending on the type of substrate used. The sandwich ELISA technique is used for many different applications including food safety testing, medication monitoring, quality control of chemicals and industrial products, and clinical diagnosis.
